(AP) ? Peyton Manning's first appearance with the Denver Broncos was a brief one.

Manning played one possession in the preseason opener against the Chicago Bears on Thursday night and got intercepted near the end zone.

In his first appearance since the Pro Bowl following the 2010 season while with Indianapolis, Manning completed 4 of 7 passes for 44 yards.

"He was remarkable," coach John Fox said. "He came back. He's worked very hard. He hasn't played football in (almost) two years. I thought it was a good first drive in these conditions. It was sloppy. It was sloppy by the receivers as far as the conditions but I thought it was a very good first outing."

On the game's first play from scrimmage, his pass attempt was broken up by Lance Briggs. Then Chicago's Geno Hayes tipped the ball near midfield, but Jacob Tamme hauled it in for a 12-yard gain.

After a short run by Willis McGahee, Manning found Eric Decker on the left for 10 yards. A 19-yarder to Decker put the ball at the 14. But on third down at the 12, Manning was picked off when Denver's Brandon Stokley deflected a pass and Major Wright dived to grab it.

NASA's Morpheus lander in fiery crash at Cape Canaveral

MIAMI | Thu Aug 9, 2012 4:11pm EDT

MIAMI (Reuters) - NASA'S Project Morpheus lander, an experimental vehicle designed with a view toward future U.S. space missions beyond Earth's orbit, crashed and burst into flames at the Kennedy Space Center in central Florida on Thursday.

During a so-called autonomous free-flight test, NASA said the vehicle lifted off the ground successfully but "then experienced a hardware component failure, which prevented it from maintaining stable flight."

No one was injured in the accident, which followed nearly a year of testing on Morpheus at the Johnson Space Center in Houston. But NASA TV footage showed the space capsule engulfed almost totally in flames after the crash, with little left to salvage.

The U.S. space agency said engineers were looking into test data to determine the exact cause of Thursday's accident, but further details were not immediately available.

"Failures such as these were anticipated prior to the test, and are part of the development process for any complex spaceflight hardware," NASA said.,

The hazard field where Morpheus had been scheduled to undergo about three months of increasingly rigorous tests was located at the north end of Kennedy's former space shuttle landing facility.

It was designed to mimic the surface of the moon, with an array of boulders, rocks, slopes and craters.

The accident came as NASA scientists were still hailing the Mars rover Curiosity's decent and landing on the "Red Planet" earlier this week as a "miracle of engineering."

Clinton aide: Nigeria military alone can't beat Islamists

ABUJA (Reuters) - The United States wants to help Nigeria fight Islamists it sees as a growing regional menace, but the country cannot rely on military might alone, an official travelling with U.S. Secretary of State Hillary Clinton said.

Clinton visited Africa's most populous nation on Thursday and met with President Goodluck Jonathan. Washington is offering support in the fight against Boko Haram, a Taliban-like group that wants to create a strict Islamic state in Nigeria's north.

She said Nigeria was "one of the most vitally important strategic partnerships in sub-Saharan Africa" in remarks at the U.S. embassy, shortly before leaving.

Africa's top oil producer is a major exporter of light crude to the United States, whose refiners favor it because it is easy to refine into motor fuel. Its proven oil and gas reserves outweigh those of the rest of sub-Saharan Africa put together.

Boko Haram has launched bomb and gun attacks on churches this year that provoked Christians, leading to deadly reprisals against Muslims. Hundreds of people have died and Washington is concerned about insecurity spreading.

"Northern Nigeria also borders Chad, it borders Cameroon, it borders Niger and we are concerned this radicalism could undermine the security of neighboring states," the senior U.S. official said.

Jonathan's critics say he relies too much on the military to defeat Boko Haram, rather than addressing northerners' grievances, such as poverty and unemployment.

"A security strategy is not enough," the official said.

Military crackdowns have mixed results - hurting Boko Haram in some areas but angering people by their heavy handedness.

Washington will offer Nigeria help with forensics, tracking of suspects and "fusing" disparate strands of police and military intelligence, the U.S. official said.

"We know all too well from our own experiences in both Iraq and Afghanistan what can happen if soldiers and police are not operating under appropriate authorities."

"We will encourage them not to use excessive force and to look at this as a ... law enforcement operation," he added.

PETROLEUM BILL

On Thursday, the Nigerian military swept through Kogi state in search of gunmen behind a massacre on Monday in which the attackers blocked exits to the Deeper Life Church in the town of Okene and fired at trapped worshippers, killing 19.

Gunmen killed three people in an attack on a mosque in the city the next day. Boko Haram have been known to attack churches and mosques.

"We have so far arrested four people including a woman. We trailed them to their hideout," Kogi state police commissioner Mohamed Katsina told Reuters by phone.

Residents complained of heavy-handed tactics in the security forces' raid, underscoring the U.S. official's concern.

"The army are raiding our houses one after the other, beating and brutalizing people," said Rahamman Bello, a resident of Adavi village on the outskirts of Okene.

"Many of our people are being arrested and molested."

Clinton was also due to address a law on oil production that has been stuck in parliament for more than five years, leaving majors such as Exxon and Chevron uncertain about the regulatory future in Nigeria.

The official said Clinton would urge a "fair and predictable environment" for oil companies in the Petroleum Industry Bill.

"If a bill comes out which appears to undermine the interest of companies, they won't invest," the official said.

Taxing the 'rich' is in the air | CalWatchDog

Aug. 8, 2012

By John Seiler

?Tax the rich? is a mantra not only in America, but globally. In California, Gov. Jerry Brown is insisting that anyone making $250,000 or more a year is ?rich? and should pay more. At the national level, President Obama also is insisting that some making $250,000 or more is ?rich? and should pay ?their fair share.?

In France, new President Hollande wants to impose a 75 percent tax on those making more than $1.2 million a year.

Hey, why ?not? The rich just rob from everybody else and splurge on yachts and Rolls Royces. They never re-invest their money in business and jobs creation. Instead, it?s governments that wisely spendsthe tax money taken from the rich, re-investing it in the education of workers, crucial research on new products and economic stimulus packages.

The Financial Times noted?on the international climate?(may be a pay wall on the link):

?Pierre Moscovici, the country?s finance minister, told Le Monde: ?This is not a punitive measure, but a patriotic measure.? The rich, he explained, are being given an opportunity to make ?an exceptional contribution? to solving France?s?financial problems?.

?The truth is that the new French government is at the extreme end of a new global trend: an international backlash against the wealthy that is reshaping politics from Europe to the US to China.?

China, of course, had long experience under Mao Zedong with punishing the rich for their sins, such as the Great Leap Forward?(1958-1961) and the Great Proletarian Cultural Revolution (1965-71). Unfortunately for the Chinese, after Mao died in 1976, his successors, especially Deng Xiaoping, unleashed greed and rich people started creating companies and exporting gizmos to the shelves of Walmart in America. But Mao still is featured in their currency, the yuan, so perhaps they can return to a time when the rich didn?t exploit the poor. The FT:

?The lifestyles of the rich and powerful is now the most sensitive and dangerous topic in Chinese politics. The?website of Bloomberg News?was recently shut down in China, apparently as punishment for the publication of an article on the family wealth of?Xi Jinping, soon to be?China?s new president.?

It?s also unfortunate that, while countries such as France and America and California are punishing the ?rich,? other countries are taking advantage of this policy. The FT:

?David Cameron, the British prime minister, has offered to roll out the red carpet for French tax exiles.??

There always are reactionaries. Both countries are part of the European Union, which makes it easy to cross borders for work and residence. Fortunately, wiser heads may prevail even there:

?But even in Britain, where the top tax rate is 45 per cent, there is a new mood of antagonism towards the rich. Even conservative politicians dare not defend bankers? pay?.

?If this new mood hardens, it could mark the end of an era of lower taxes, deregulation and rising inequality that began in the late 1970s, with the rise of Margaret Thatcher and Ronald Reagan in the west and of Deng Xiaoping in China When Lady Thatcher came to office in 1979, Britain?s top tax rate was 83 per cent. She cut it, first to 60 per cent and then to 40 per cent ? where it stayed until the financial crisis. Reagan inherited a top income tax rate of 70 per cent and cut it to 50 per cent and finally to 28 per cent. In China, Deng Xiaoping captured the spirit of the times when he remarked: ?To get rich is glorious.??

The new theme is: ?To be taxed is glorious.?

Currently in California, the ?rich? pay a top federal income tax rate of 35 percent and a top California state tax rate of 10.3 percent. Total: 45.3 percent.

If all the tax increases go through, those numbers would go, respectively, to 38.6 percent and 13.3 percent. Total: 51.9 percent.

Actually, an even better idea would be to restore the 91 percent federal level from the 1950s. Combined with the 13.3 percent California top tax rate, if Brown?s tax increase is passed, the top rate here then would be 104.3 percent.

A tax rate of 104.3 percent would benefit the rich the most because their money would be ploughed back into the economy on the construction of infrastructure projects such as the California High-Speed Rail Authority, thus generating more wealth for them and everybody else.

The 104.3 percent income tax rate: It?s an idea whose time has come.

Russian sect found after nearly a decade

MOSCOW (Reuters) - Seventy members of an Islamist sect who have been living in an underground bunker without heat or sunlight for nearly a decade have been discovered living on the outskirts of the city of Kazan in Russia, local media reported.

The sect members included 20 children, the youngest of whom had just turned 18 months. Many of them were born underground and had never seen daylight until the prosecutors discovered their dwelling on August 1 and sent them for health checks.

A 17-year-old girl turned out to be pregnant.

Religion was suppressed in the Soviet Union which collapsed in 1991, prompting various cults and sects to flourish in the vacuum that opened up.

The group - known as the "Fayzarahmanist" sect - was named after its 83-year-old organiser Fayzrahman Satarov, who declared himself a prophet and his house an independent Islamic state, according to a report by state TV channel Vesti.

Satarov was described as a former deputy to a Sunni Islamic cleric in the 1970s. His followers were encouraged to read his manuscripts and most were banned from leaving their eight-storey underground bunker which had been dug in the basement of a building, Vesti said.

Prosecutors have opened a criminal investigation into the sect and have said it will be disbanded if it continues its illegal activities, such as stopping its members from seeking medical assistance or education.

No arrests have been made although police are likely to look into suspicions that some of the children were being abused. A court will decide whether the children will be allowed to stay with their parents.

Kazan is located 800 km (497 miles) east of Moscow in Tatarstan, a majority Muslim internal Russian republic.

Sob: Thief 4 Sneaking Onto Next Gen Consoles? | Rock, Paper ...

By John Walker on August 8th, 2012 at 6:00 pm.

The release year.

While Thief 4 has never had anything approaching a release date, following the ripe success of Deus Ex 3, and its having been in development for over four years, it was tempting to think it might be coming nearer. Well, forget that, because a rumour in the latest Official Xbox Magazine ? and further validated by CVG ? suggests that they?re now aiming for it to be a next-next-gen console release. And that means it?d be Christmas 2013 at the very, very earliest.

Rumours have flown every whichway about the project, mostly because Square Enix and Eidos Montreal have been so damned silent about it. That tends to indicate a project that?s in trouble, as a rule ? one that keeps receiving do-overs, reaching certain milestones and not being considered good enough. Of course, there?s no evidence at all that that?s the case here. But things have been odd, with a completed trailer from December last year never getting released, and any footage that?s appeared from it having since vanished from the internet, entire blogs deleted. Kotaku also lists the many developers who have left the project in the last year or so, but bear in mind it?s not unusual for people to move on to a new project after three or four years. And they also spot that level designer, Fabien Morisson, pointed out on his LinkedIn that he?d designed the last level of the game.

Deciding to make the game for the next generation of consoles could mean only small or absolutely massive changes. If their engine is scalable, and just needed better tech to be better realised, then it?ll just be a case of getting to grips with the debug prototype consoles they?ll have by now, and porting it across. If they?re after new tech because the version they?ve made just can?t reach far enough, then it could mean a massive overhaul. Either way, it means a PC version of the game will inevitably get stuck waiting for Microsoft and Sony to release their next boxes, both rumoured to be for Christmas next year. Then you have to wonder if Square are going to be willing to risk being launch titles, going out to the most limited audience possible, or wait a few months for the consoles to be in enough homes. That would put the game somewhere into 2014.

Which is all to say, don?t hold your breath (while crouching in a dark corner) for the game.

We approached Square Enix for comment, but they had none.

Rapid Rise Seen in Antipsychotic Prescriptions for Children and ...

Antipsychotic treatment has increased rapidly among young people in the United States, with much of the increase coming from prescriptions for disruptive behavior disorders such as attention deficit/hyperactivity disorder (ADHD), Reuters reports.

In the Archives of General Psychiatry, the researchers report that antipsychotic drugs are prescribed during almost one in three visits children and teenagers make to psychiatrists in the United States, an increase from one in 11 in the 1990s.

Most of the antipsychotics are not prescribed for conditions approved by the Food and Drug Administration (FDA). In children and teenagers, antipsychotics are indicated for irritability associated with autistic disorder, tics and vocal utterances of Tourette syndrome and bipolar mania, and schizophrenia.

Researcher Dr. Mark Olfson, of Columbia University in New York, found that about 90 percent of antipsychotic prescriptions written during office visits between 2005 and 2009 were ?off label,? or prescribed for a condition that has not been approved by the FDA. The article notes the effectiveness of antipsychotic drugs for ADHD is uncertain. The drugs are associated with weight gain and diabetes.

?There is very little question as to whether these drugs are being prescribed in kids much more than they used to,? Olfson told Reuters. He added he hopes parents will ask doctors more questions about antipsychotics, and whether there are other treatment options, such as parent management training, to reduce aggressive and disruptive behavior in children.

Risk of stroke from cardiac catheterizations

MAYWOOD, Il. -- When a patient undergoes a cardiac catheterization procedure such as a balloon angioplasty, there's a slight risk of a stroke or other neurological complication.

While the risk is extremely small, neurologists nevertheless may expect to see catheterization-induced complications because so many procedures are performed, Loyola neurologists write in the journal MedLink Neurology.

Cardiac catheterizations include diagnostic angiograms, balloon angioplasties and stent placements. More than 1.4 million procedures are successfully performed each year. Cardiac catheterizations, like all medical treatments, carry some degree of risk. But because the risk is low, neurologists rarely see patients who experience neurological complications. The purpose of the MedLink article is to raise awareness of the risks and to list treatment options for when complications do occur.

The procedure involves inserting a catheter (thin tube) in the groin or arm and guiding it to the heart. In rare cases, debris can be knocked loose from blood vessel walls, travel to the brain and trigger a stroke or transient ischemic attack (mini stroke). Tiny bubbles released from the catheter also can trigger a stroke or transient ischemic attack. And bleeding in the groin or arm where the catheter is inserted can cause peripheral nerve damage.

However, the risk is slight. And with the use of more refined techniques and smaller and softer catheters, the risk is getting even smaller, said H. Steven Block, MD, first author of the review article.

"We want to be careful to not scare people who need a cardiac catheterization from getting this beneficial procedure," Block said.

Indeed, because the incidence is so low, it is difficult to perform randomized clinical trials to determine the best treatment for catheterization-induced neurologic complications, the authors write.

"Cardiac catheterization is a very safe procedure," Block added. "A lot of neurologists may encounter neurologic complications only once or twice during their careers. But we would like to raise awareness and knowledge, so they are better prepared when a case does happen."

Block is a mid-career neurology fellow at Loyola. Co-authors are Loyola neurologists Sarkis Morales-Vidal, MD; Alejandro Hornik, MD; and Jose Biller, MD. Biller is chair of Loyola's Department of Neurology.

Opinion: Comparing Mexican, U.S. Electoral Systems: Which Is the Banana Republic?

On July 1, Mexicans cast their ballots for president, Congress, and state governors. This marks the third presidential election since the nation's entire electoral system was reformed beginning in 1990, and as in the two earlier elections, the outcomes have been accepted. News reports described the electoral process having taken place with limited problems for voters.

Mexico?s next president will be Enrique Pena Nieto, candidate of the Institutional Revolution Party, known as PRI, who has officially won with 38 percent of the vote. The runner-up this time as well as in 2006 was Andres Manuel Lopez Obrador of? the Democratic Revolution Party, or PRD, with 32 percent while National Action Party candidate Josefina Vazquez Mota placed third with 25 percent. These results closely aligned with final official preelection polling announced June 27.

In 2006, Mexico's presidential election ended in a virtual tie, and it took two months to determine the outcome. Despite cries of fraud from the Left, the outcome gave National Action Party candidate Felipe Calderon a 243,934 vote margin of victory. Protests similar to Occupy Wall Street took place throughout the country, but once the Federal Electoral Commission declared Calderon the winner, the verdict was respected and accepted by all but a few Mexicans. If one wants credible, simple, and sensible elections, one need only look to Mexico as a shining example.

Bear in mind, that for 75 years, Mexico had one of the most corrupt and fraudulent electoral systems among so-called democratic governments. All this changed in the early 1990s when Mexico established a nongovernment organization, the Federal Electoral Institute, a totally nonpartisan institution respected by all legitimate parties. Today, its system is a model to emerging democracies such as Haiti and Iraq. The practice of Iraqis dipping their thumbs in blue ink upon voting was adopted from the Mexican system. After voting, Mexicans dip their thumbs in purple ink.

Compared with the Mexican electoral system, the American system is woefully inefficient and open to fraud. Our current system is actually a composite of 50 systems that vary from state to state. Some require photo identification, others do not. Some allow for no-fault absentee voting. Others do not. Some allow for early voting or Internet voting. Currently, 16 states have enacted a photo-ID mandate. Fifteen states, including Ohio, require voters to show some form of personal identification such as a utility bill or a bank statement.

Mexicans are very scrupulous about documentation for official purposes, so one must present required documents to register to vote. There's no such thing as ?motor-voter registration? as is done here; a new Mexican voter has to take a birth certificate and proof of residency to an IFE office to register.

Once the application is complete, the newly registered voter is photographed and fingerprinted. With this, an official voter ID card is created containing picture, thumbprint, and voter encrypted data. Registration must be renewed every 12 years. It takes about two weeks for the card to be created, at which time the voter must return to the office personally sign for, and collect it. It is not mailed. The Federal Electoral Voting Card cannot be tampered with or altered and is the gold standard for identification purposes.

To allow for the greatest possible access to voting, Election Day is always on Sunday in Mexico. Everyone votes on the same day, no exceptions. Absentee voting? It doesn't exis, because of the high potential for fraud. ?There are no provisional ballots and no possibility of ?hanging chads.? Polls open and close at the same time in all parts of the country. Beginning in 2006, voting was extended to voters outside of Mexico, particularly to accommodate the millions of registered voters in the U.S. The poor and disadvantaged are especially encouraged to register and vote.

Nearly 80 million Mexicans were eligible to vote in the July 1 election, and an estimated 62 percent actually voted, more than the norm in American presidential elections. Americans can only hope that our presidential election in November will be as noncontroversial as Mexico?s and that the outcome will be respected. The Mexican system is not perfect and has various shortcomings which can and should be addressed.

The Mexican electoral system versus the American electoral system: Which is the banana republic?

Making Money Blogging In The UK For Your Business

This article has been viewed 35 times.

It doesn?t take quite a lot of research to search out out that this present day, just about each business of any real size has developed some form of internet presence. Now, for many companies, that may imply little greater than a web based enterprise card that can be used to get the phone quantity and retailer location of the enterprise into the mind of the prospective customer. However on this new century, the idea of getting a business with out a corresponding internet page to help it?s pretty much out of the question.

However should you look at the two enterprise worlds, the internet business surroundings and that outdoors of our on-line world, there are some pretty large differences. While many companies like bookstores or live performance ticket promoters have discovered to build what is perhaps seen as parallel universes during which their enterprise operations are simply as sophisticated on-line as outside of cyberspace, different businesses have simply not discovered that balance.

But because the legitimacy of the internet as a valid market and business software becomes extra understood, increasingly businesses are studying that cyberspace can change into one other valuable part of an general marketing plan that drives enterprise to the store shelves immediately from their web internet presence.

So simply as that billboard or newspaper coupon program are simply as much part of the businesses company plan, that on-line effort on the market in on the company web page can turn out to be an important a part of the stores operation so much so that the shop supervisor will come to rely on the gross sales driven by the internet. To that store manager they will search for our on-line world on aisle five as an important a part of their plan for business success.

There?s a systematic course of that companies go through to use the web as a way of capturing web site visitors and turning it into store traffic. Make no mistake, there may be one principle that ought to seem evident however is the important thing to turning cyber guests to in store customers and that is that ? Web Shoppers are People Too!

When a businessperson looks at those unusual web traffic reports that show that they site has X variety of ?hits? and that Z number of web browsers went to Y variety of web pages, all of that cyberspace mumbo jumbo simply signifies that X variety of PEOPLE had been in your web page and checked out Y variety of merchandise or internet page commercials or services. And people PEOPLE are the same residing and respiratory people who will walk within the front door of your store and purchase services and products from you.

All we need to do is devise strategies to drive these internet consumers off of their computer systems and into the companies retail operations. And increasingly you might be seeing a businesses development of web promotions that are geared to place the customers ft down within the retail space. Some great strategies for doing which can be.

* On-line coupons that can be redeemed only within the retail store.
* Online sales that may be picked up in the store. Many web shoppers would possibly prefer to have the product shipped to them. So you will have to ?sweeten the pot? by making delivery fees out of the question or by including a promotion if the customer picks up his or her buy in person.
* Contests. Need we say extra?
* By selling particular events that will occur in the store. You?ll be able to stage a significant our on-line world promotional marketing campaign for a guide signing of an author or celeb that will occur live at the retailer itself. The costs of the promotion and having the in retailer event shall be offset by the increased sales.

If your website online routinely uses promotions that end in constructive incentives to the customer to come to the shop, earlier than long a buyer base of loyal customers will get used to first going online to see what this week?s large deal is and then going to the shop to money in. That type of ongoing momentum is what makes such a synergy such a success and what makes even customers come to your retail outlet and look for ?cyberspace on aisle five.?

Irony seen through the eye of MRI

ScienceDaily (Aug. 3, 2012) ? In the cognitive sciences, the capacity to interpret the intentions of others is called "Theory of Mind" (ToM). This faculty is involved in the understanding of language, in particular by bridging the gap between the meaning of the words that make up a statement and the meaning of the statement as a whole.

In recent years, researchers have identified the neural network dedicated to ToM, but no one had yet demonstrated that this set of neurons is specifically activated by the process of understanding of an utterance. This has now been accomplished: a team from L2C2 (Laboratoire sur le Langage, le Cerveau et la Cognition, Laboratory on Language, the Brain and Cognition, CNRS / Universit? Claude Bernard-Lyon 1) has shown that the activation of the ToM neural network increases when an individual is reacting to ironic statements.

Published in Neuroimage, these findings represent an important breakthrough in the study of Theory of Mind and linguistics, shedding light on the mechanisms involved in interpersonal communication.

In our communications with others, we are constantly thinking beyond the basic meaning of words. For example, if asked, "Do you have the time?" one would not simply reply, "Yes." The gap between what is said and what it means is the focus of a branch of linguistics called pragmatics. In this science, "Theory of Mind" (ToM) gives listeners the capacity to fill this gap. In order to decipher the meaning and intentions hidden behind what is said, even in the most casual conversation, ToM relies on a variety of verbal and non-verbal elements: the words used, their context, intonation, "body language," etc.

Within the past 10 years, researchers in cognitive neuroscience have identified a neural network dedicated to ToM that includes specific areas of the brain: the right and left temporal parietal junctions, the medial prefrontal cortex and the precuneus. To identify this network, the researchers relied primarily on non-verbal tasks based on the observation of others' behavior[1]. Today, researchers at L2C2 (Laboratoire sur le Langage, le Cerveau et la Cognition, Laboratory on Language, the Brain and Cognition, CNRS / Universit? Claude Bernard-Lyon 1) have established, for the first time, the link between this neural network and the processing of implicit meanings.

To identify this link, the team focused their attention on irony. An ironic statement usually means the opposite of what is said. In order to detect irony in a statement, the mechanisms of ToM must be brought into play. In their experiment, the researchers prepared 20 short narratives in two versions, one literal and one ironic. Each story contained a key sentence that, depending on the version, yielded an ironic or literal meaning. For example, in one of the stories an opera singer exclaims after a premiere, "Tonight we gave a superb performance." Depending on whether the performance was in fact very bad or very good, the statement is or is not ironic.

The team then carried out functional magnetic resonance imaging (fMRI) analyses on 20 participants who were asked to read 18 of the stories, chosen at random, in either their ironic or literal version. The participants were not aware that the test concerned the perception of irony. The researchers had predicted that the participants' ToM neural networks would show increased activity in reaction to the ironic sentences, and that was precisely what they observed: as each key sentence was read, the network activity was greater when the statement was ironic. This shows that this network is directly involved in the processes of understanding irony, and, more generally, in the comprehension of language.

Next, the L2C2 researchers hope to expand their research on the ToM network in order to determine, for example, whether test participants would be able to perceive irony if this network were artificially inactivated.

Note:

[1] For example, Gr?zes, Frith & Passingham (J. Neuroscience, 2004) showed a series of short (3.5 second) films in which actors came into a room and lifted boxes. Some of the actors were instructed to act as though the boxes were heavier (or lighter) than they actually were. Having thus set up deceptive situations, the experimenters asked the participants to determine if they had or had not been deceived by the actors in the films. The films containing feigned actions elicited increased activity in the rTPJ (right temporal parietal junction) compared with those containing unfeigned actions.

Researchers discovered a molecule that converts stem cells into heart cells, which could be used to replace diseased or damaged tissue in heart disease patients

LA JOLLA, Calif., August 2, 2012 For years, scientists have been looking for a good source of heart cells that can be used to study cardiac function in the lab, or perhaps even to replace diseased or damaged tissue in heart disease patients. To do this, many are looking to stem cells. Researchers at Sanford-Burnham Medical Research Institute (Sanford-Burnham), the Human BioMolecular Research Institute, and ChemRegen, Inc. have been searching for molecules that convert stem cells to heart cells for about eight yearsand now they've found one. Writing in the August 3 issue of Cell Stem Cell, the team describes how they sifted through a large collection of drug-like chemicals and uncovered ITD-1, a molecule that can be used to generate unlimited numbers of new heart cells from stem cells.

"Heart disease is the leading cause of death in this country. Because we can't replace lost cardiac muscle, the condition irreversibly leads to a decline in heart function and ultimately death. The only way to effectively replace lost heart muscle cellscalled cardiomyocytesis to transplant the entire heart," said Mark Mercola, Ph.D., director of Sanford-Burnham's Muscle Development and Regeneration Program and senior author of the study. "Using a drug to create new heart muscle from stem cells would be far more appealing than heart transplantation."

Searching for a needle in a haystack

Stem cells are important because they do two unique things1) self-renew, producing more stem cells and 2) differentiate, becoming other, more specialized cell types. To obtain a large number of a certain cell type, such as heart cells, the hard part is figuring out the signals that direct them to become the desired cell type.

Mercola's group has been hunting for heart-inducing signals for 15 yearsin embryos and in stem cells. To find a synthetic molecule that might one day lead to a drug therapy to regenerate the heart, they joined forces with a team of medicinal chemists at the Human BioMolecular Research Institute led by John Cashman, Ph.D. With funding from the California Institute for Regenerative Medicine, they used sophisticated robotic technology to methodically test a large collection of drug-like chemicals, looking for that needle in a haystack that, when added to stem cells, results in cardiomyocytes. The winning compound was ITD-1.

Therapeutic applications

There's no shortage of therapeutic possibilities for ITD-1. "This particular molecule could be useful to enhance stem cell differentiation in a damaged heart," explained Erik Willems, Ph.D., postdoctoral researcher in Mercola's lab and first author of the study. "At some point, it could become the basis for a new therapeutic drug for cardiovascular diseaseone that would likely limit scar spreading in heart failure and promote new muscle formation."

Mercola, Willems, and Cashman are now working with San Diego biotech company ChemRegen, Inc. to further develop ITD-1 into a drug that one day might be used to treat patients.

More scientific detail

The researchers discovered that ITD-1 blocks a cellular process known as TGF? signaling. TGF? (short for transforming growth factor-?) is a protein produced by one cell type to influence others' behaviors, such as proliferation, scarring, and even stem cell differentiation. TGF? works from outside the cell, binding to a receptor on the surface of a responding cell to initiate an intracellular signaling cascade that causes genes to be switched on or off, ultimately altering cellular behaviorin this case making heart muscle.

ITD-1 triggers degradation of the TGF? receptor, thus inhibiting the whole process. With TGF? signaling turned off, stem cells are set on a course toward cardiogenesis. ITD-1 is the first selective inhibitor of TGF?, meaning that it might also have applications in many other processes controlled by TGF?.
